What causes the formation of Lewy bodies?

Lewy bodies consist of misfolded forms of a protein called alpha-synuclein; their exact cause of formation is yet to be deciphered but several factors including genetic variants, protein malfunction, oxidative stress, inflammation, age-associated changes (including impaired protein transport and degradation as well as mitochondrial dysfunction) may play a role.
